Growth Hormone and Growth Hormone Releasing Hormone: Understanding the Mechanisms for the Decline of Growth Hormone with Age

It is established across species that growth hormone secretion declines with age. My research has concentrated on understanding the relationship between changes in body composition, age, gonadal steroids and growth hormone secretion. Low levels of growth hormone are associated with increased adiposity, particularly in visceral fat. This increased visceral fat is associated with increased cardiovascular risk and reduced life expectancy and morbidity. We are performing clinical studies to understand the mechanism by which the decline of growth hormone occurs in aging. Our primary hypothesis is that this is due to a progressive decline in GHRH secretion from the hypothalamus associated with neuronal cell loss in aging brain. A second series of studies are examining the possibility of restoring growth hormone secretion to that seen in young individuals either by administering GHRH or by administering growth hormone secretagogues. We also have a basic laboratory which is oriented towards understanding the signal transduction pathway for growth hormone action and a separate laboratory dealing with the molecular biology and physiology ofthe regulation of the receptor for the growth hormone releasing hormone. Our major effort is to establish whether enhanced growth hormone secretion in aging individuals is able to improve fitness and restore body composition to that found in younger individuals. It is hoped that this will improve well-being, reduce morbidity and improve life expectancy.
